目的:探讨庆大霉素新剂型—水杨酸庆大霉素的耳毒性作用。方法:将实验动物随机分为五组,即庆大霉素(GM)组;水杨酸庆大霉素(SAGM)组;庆大霉素+水杨酸钠(GM+NaSA)组;水杨酸钠(NaSA)组和对照(C)组。实验前后检测动物ABR反应阈;耳蜗铺片观察毛细胞缺失程度;电镜观察毛细胞超微结构;并检测血清和外淋巴铁及肾功能。结果:SAGM组ABR阈移较GM组小(P<0.05),各频率和时间点与(GM+NaSA组无明显差异(P>0.05)。耳蜗铺片和电镜观察显示,SAGM组毛细胞损失程度和超徽结构变化比较GM组明显减轻。各组间血清铁和外淋巴铁无显著差异,SAGM对肾功能无明量影响。结论:新剂型水杨酸庆大霉素的耳毒性较硫酸庆大霉素明显减轻。
Objective: To investigate the ototoxic effect of gentamicin salicylate, a new dosage form of gentamicin. Methods: The experimental animals were randomly divided into five groups: gentamycin (GM) group, gentamicin salicylate (SAGM) group, gentamycin + sodium salicylate (GM + NaSA) Sodium salicylate (NaSA) group and control (C) group. The ABR threshold of the animals was measured before and after the experiment. The degree of hair loss was observed by cochlear mapping. The ultrastructure of the hair cells was observed by electron microscope. The serum and extra-lymphoid iron and renal function were detected. Results: Compared with GM group, the threshold of ABR threshold in SAGM group was lower than that in GM group (P <0.05), and there was no significant difference between the two groups (P> 0.05) Degree and ultrastructure changes of GM group were significantly reduced.The serum iron and extra-lymph node iron between the groups showed no significant difference, SAGM no significant effect on renal function.Conclusion: The new formulation of gentamicin salicylate ototoxicity than sulfuric acid Gentamycin significantly reduced.
