摘要目的明确MR扫描时不同强度场强(1、1.5、3、7 T)和对比剂对病人DNA双链断裂(DSB)的作用。材料与方法该活体实验通过机构伦理委员会批准,病人均签署知情同意书。在进行MRI检查前及检查后的5、30 min,直接从外周血样本中分离出单核细胞以分析DNA的DSB水平。采用γH2AX免疫荧光染色,自动数字显微镜定量分析DSB。MR小组由43例病人组成(女22例,男21例;年龄20~77岁,平均46.1岁),并依据施加的场强和对比剂分为不同亚组。此外,10例进行CT平扫或CT增强扫描的病人作为阳性对照
Abstract Objective To clarify the effect of different intensities (1, 1.5, 3, 7 T) and contrast agent on DNA double-strand break (DSB) in MR scan. Materials and Methods The in vivo experiments were approved by institutional ethics committee and patients signed informed consent forms. Mononuclear cells were isolated directly from peripheral blood samples to analyze DNA DSB levels before and at 5,30 min after MRI examination. ΓH2AX immunofluorescence staining, automatic digital microscope quantitative analysis of DSB. The MR subgroup consisted of 43 patients (22 females, 21 males; 20 to 77 years of age, mean 46.1 years) and were divided into subgroups based on applied field strength and contrast agent.
