目的:研究三七皂甙对大鼠慢性缺氧性肺动脉高压的作用。方法:采用间断常压缺氧法制备大鼠慢性缺氧性肺动脉高压模型,腹腔注射三七皂甙(25,50和100mg·kg-1·d-1),连续给药21天。结果:三七皂甙能产生剂量依赖性地抑制慢性缺氧性肺动脉高压的作用,其抑制率分别为15.9%、43.9%和73.4%,并能明显抑制右心室肥厚,其抑制率分别为22.1%、55.6%和83.3%,而对颈动脉血压无明显的影响。三七皂甙(200g·L-1)能明显抑制血小板生长因子所诱导的离体肺动脉平滑肌细胞DNA的合成。结论:三七皂甙有抑制慢性缺氧性肺动脉高压的作用,可能是通过抑制血小板生长因子促肺动脉平滑肌细胞增殖而抑制肺血管结构重建来实现的。
Objective: To study the effect of panax notoginseng on chronic hypoxic pulmonary hypertension in rats. METHODS: Rats with chronic hypoxic pulmonary hypertension were prepared by intermittent atmospheric hypoxia and were given intraperitoneal injection of panax notoginseng saponins (25, 50 and 100 mg·kg-1·d-1) for 21 consecutive days. RESULTS: Panax Notoginseng could inhibit the effects of chronic hypoxic pulmonary hypertension in a dose-dependent manner. The inhibitory rates were notably 15.9%, 43.9% and 73.4%, respectively, and significantly inhibited right ventricular hypertrophy. The inhibition rates were 22.1%, 55.6%, and 83.3%, respectively, but no significant effect on carotid blood pressure. Panax notoginseng saponins (200g·L-1) can significantly inhibit the synthesis of DNA isolated from pulmonary artery smooth muscle cells induced by platelet growth factor. Conclusion: Panax Notoginseng has the effect of inhibiting chronic hypoxic pulmonary hypertension. It may be achieved by inhibiting the proliferation of pulmonary artery smooth muscle cells and inhibiting the remodeling of pulmonary vascular structure.
