目的:探讨磷酸酯酶2B(PP-2B)的抑制对神经瘤细胞代谢及细胞骨架结构的影响.方法:神经瘤细胞与PP-2B特异性抑制剂共培养,用结晶紫染色和MTT法测定细胞活性,光学显微镜检测细胞形态,免疫细胞染色法检测细胞骨架及PP-2B表达活性.结果:抑制PP-2B可导致细胞代谢明显降低,细胞成活数显著减少;细胞突起消失;微管受损.结论:PP-2B缺陷导致神经瘤细胞代谢和结构双重异常.“,”Aim: To explore the effect of protein phosphatase 2B(PP-2B)inhibition on the metabolism and structure of neuro-blastema cell. Methods: Neuroblastoma cell co-culture with PP-2B inhibitor (trifluoper-azine), crystal violet staining and MTT were used to evaluate the cell viability and activity, phase contract microscopy and immunocytochemistry were applied separately to detect the cytoskeleton and the expression of PP-2B. Results: The inhibition of PP-2B by trifluoperazine leaded to a markedly decreased metabolism and a significantly increased cell death. A differentially damaged cell process and the lesion of microtubule system could also be seen by treatment of the cell with different concentration of the inhibitor. Conclusion: PP-2B may play a key role in maintaining normal metabolism and integrity of microtubules in neuroblastoma cells and the deficiency of PP-2B causes the cell damage both metabolically and structurally.
