目的探查甘丙肽能否促进大鼠脂肪细胞GluT4膜位移。方法用3 H葡萄糖测3T3L1脂肪细胞葡萄糖(2DG)摄取率;大鼠分4组:安静和运动对照组注射生理盐水,安静和运动用药组注射甘丙肽拮抗剂M35。两运动组大鼠游泳20d,60min/d。结果甘丙肽增加葡萄糖摄取;两用药组大鼠正糖钳的葡萄糖输注速率及脂肪细胞GluT4mRNA水平均下降,细胞外膜和总膜GluT4浓度降低,且外膜与总膜GluT4比值低于对照组。结论甘丙肽可促进GluT4膜转位而增强胰岛素敏感性。
Objective To investigate if galanin can promote GluT4 membrane migration in rat adipocytes. Methods Glucose uptake rate of 3T3L1 adipocytes was measured by 3 H glucose. The rats were divided into 4 groups: normal and exercise control groups were injected with normal saline, and the rats in the quiet and exercise groups were injected with galanin antagonist M35. Rats in both exercise groups swam for 20d and 60min / d. Results Galanin increased glucose uptake. Glucose infusion rate and GluT4 mRNA level were decreased in positive glucose clamp group and GluT4 concentration in outer membrane and total membrane, and ratio of outer membrane to total membrane GluT4 was lower than that of control group. Conclusion Galanin can promote GluT4 membrane translocation and enhance insulin sensitivity.
