为进一步探索人精子鞘糖脂(hsGSL)在精卵识别中的重要作用,本文采用了Ladisch分配,Vance法,高效薄板层析及薄层扫描等方法对hsGSL进行了纯化、鉴定和定量.人精了神经节苷脂(hsGls)中脂结合唾液酸的含量为0.18±0.05μg/10~8个精子细胞;人精子中性鞘糖脂(hsNGSL)中鞘氨醇的含量为0.64±0.10μg/10~8个精子细胞.hsGls主要由GM3(26.93±8.82%),GM2(15.60±3.35%),GD3(28.94±4.02%),GDla(18.56±3.42%),及GTlb(9.87±1.56%)五种组分组成.hsNGSL主要由CMH(10.95±4.41%),CDH(16.09±2.0%),CTH-1(32.73±0.09%),CTH-2(20.95±3.05%),CQH(11.24±1.74%)及CPH(8.04±0.57%)六种组分组成.
In order to further explore the important role of hsGSL in sperm-egg recognition, hsGSL was purified, identified and quantified by Ladisch distribution, Vance method, high-performance thin-layer chromatography and TLC. The content of lipid-bound sialic acid in hsGls was 0.18 ± 0.05μg / 10-8 sperm cells. The content of sphingosine in human sperm sphingosine (hsNGSL) was 0.64 ± 0.10μg / 10-8 sperm cells.HsGls mainly consists of GM3 (26.93 ± 8.82%), GM2 (15.60 ± 3.35%), GD3 (28.94 ± 4.02%), GDla (18.56 ± 3.42%) and GTlb (9.87 ± 1.56% ) .HsNGSL was mainly composed of CMH (10.95 ± 4.41%), CDH (16.09 ± 2.0%), CTH-1 (32.73 ± 0.09%), CTH-2 (20.95 ± 3.05%), CQH 1.74%) and CPH (8.04 ± 0.57%) six components.
