借助高效液相色谱-高分辨质谱(HPLC-DAD/ESI-Q-TOF-MS)联用技术,对两面针中的生物碱进行初步分析。采用汉邦C18色谱柱,以0.1%甲酸-乙腈和0.1%甲酸-水为流动相,梯度洗脱,经柱后分流分别经DAD检测器及ESI-Q-TOF-MS正离子模式采集。根据获得的母离子和碎片离子精确相对分子质量信息,结合文献,从两面针中鉴定分析了48个成分,其中,6个成分为新化合物。该方法分离度良好、灵敏度高,可用于两面针药材的定性分析,并有助于对该药材的质量控制及深入研究。
Alkaloids in two-sided needles were analyzed by HPLC-DAD / ESI-Q-TOF-MS. Using Hanbang C18 column, mobile phase was eluted with 0.1% formic acid-acetonitrile and 0.1% formic acid-water as gradient eluent and separated by column after DAD detector and ESI-Q-TOF-MS. According to the accurate relative molecular mass information of precursor ions and fragment ions obtained, 48 components were identified and identified from the two-sided needle with reference to the literature, of which 6 were new compounds. The method has good resolution and high sensitivity, which can be used for the qualitative analysis of medicinal materials of two-sided needles and contributes to the quality control and further study of the medicinal materials.
