目的 探讨γ-分泌酶抑制剂中的DAPT对结核病(TB)患者外周血单个核细胞(PBMCs)的凋亡和增殖作用.方法 收集TB患者、潜伏感染者(LTBI)和结核菌素皮试实验阴性(TST-)健康者外周血标本各30例,淋巴细胞分离液分离PBMCs,应用不同浓度DAPT(0、2.5、5、10和20μmol/L)处理PBMCs,应用显微镜计数PBMCs数量,流式细胞术法测定细胞凋亡和细胞增殖情况.结果 与DMSO组比较,TST-和LTBI组细胞数均随着DAPT浓度增加剂量依赖性降低,而TB患者在DAPT为10μmol/L时细胞数量减少明显,凋亡明显增加,差异有统计学意义(P0.05).结论 DAPT可以增加结核病患者PBMCs凋亡,在DAPT浓度为10μmol/L时凋亡最明显,但不影响细胞增殖.“,”Objective To study the apoptotic and proliferation effect of gamma-secretase inhibitor(DAPT)on peripheral blood mononuclear cells (PBMCs) of patients with tuberculosis (TB). Methods Peripheral blood specimens were collected from TB patients,latent tuberculous infection(LTBI)and tuberculin skin test negative(TST-)individuals;each group had 30 cases. PBMCs were separated by ficoll-hypaque solution;PBMCs were treated by different concentrations(0, 2.5,5,10 and 20 μmol/L)of DAPT;the number of PBMCs was counted by microscope;cell apoptosis and proliferation were determined by flow cytometry method. Results Compared with DMSO group ,PBMCs number was reduced in a dose dependent in TST negative and LTBI groups,while when DAPT was 10μmol/L,the PBM cells were decreased significantly and apoptotic cells were obvious in TB patients(P0.05). Conclusion DAPT could increase cell apoptosis at 10μmol/L DAPT,but not affect proliferation in TB patients.
