目的探讨黄芪当归合剂降脂及减少肾小球细胞外基质沉积的作用是否与肾脏转化生长因子(TGF)β1表达的改变有关。方法用嘌呤霉素致肾病综合征大鼠,随机分肾病对照、中药、西药治疗三组。用免疫组化及原位杂交观察肾脏TGFβ1表达及细胞外基质(ECM)沉积。以计算机图象分析进行半定量比较。结果正常组,可见TGFβ1蛋白及mRNA于肾小管上皮细胞微弱表达,肾小球系膜区TGFβ1mRNA弱表达。其余各组TGFβ1蛋白表达位于肾小管上皮细胞,mRNA表达位于肾小球系膜细胞及肾小管上皮细胞,而后者表达更明显。两治疗组TGFβ1表达较肾病组明显降低(P<0.001),两治疗组间比较,无显著性差异。两治疗组细胞外基质沉积均明显少于肾病对照组。结论黄芪当归合剂及HMGCoA还原酶抑制剂对肾病综合征鼠能明显减少肾小球细胞外基质沉积,同时使TGFβ1表达的下调。提示黄芪当归合剂可能通过这一下调作用减少肾小球细胞外基质的沉积
Objective To investigate whether the effect of Astragalus and Angelica mixture on lipid-lowering and glomerular extracellular matrix deposition is related to the change of transforming growth factor (TGF) β1 in the kidney. Methods Puromycin induced nephrotic syndrome rats were randomly divided into three groups: control of kidney disease, Chinese medicine and Western medicine. Kidney TGFβ1 expression and extracellular matrix (ECM) deposition were observed by immunohistochemistry and in situ hybridization. Semi-quantitative comparison with computer image analysis. Results In the normal group, the expression of TGFβ1 protein and mRNA in renal tubular epithelial cells was weak and the expression of TGFβ1 mRNA in the glomerular mesangial area was weak. The other groups of TGFβ1 protein expression in renal tubular epithelial cells, mRNA expression in glomerular mesangial cells and renal tubular epithelial cells, while the latter expression is more obvious. TGFβ1 expression in the two treatment groups was significantly lower than that in the nephropathy group (P <0.001). There was no significant difference between the two treatment groups. The extracellular matrix deposition in both treatment groups was significantly less than that in the control group. Conclusion Astragalus and Angelica mixture and HMGCoA reductase inhibitors can significantly reduce glomerular extracellular matrix deposition and down-regulate the expression of TGFβ1 in rats with nephrotic syndrome. Tip Astragalus Angelica mixture may be through this down-regulation of glomerular extracellular matrix deposition
