噬菌体抗体库技术的出现标志着抗体技术从多克隆抗体、单克隆抗体进入了基因工程抗体的时代。这一技术具有省时、省力,筛选容量大,可直接得到抗体基因,便于构建各种基因工程抗体及在原核系统中进行表达的特点。尤其是可以不经免疫而获得针对任何抗原的抗体以及人源抗体的获得,具有划时代的意义。获得预期的特异性抗体的关键是构建的库具有足够大的容量和多样性以及强有力的筛选手段。目前从方法上仍需不断改进和完善,本文就目前抗体库构建中如何增加库容量和多样性,以及筛选方法做一综述。
The emergence of phage antibody library technology marks the era of antibody technology from polyclonal antibodies, monoclonal antibodies into the genetic engineering of antibodies. This technique has the characteristics of time-saving, labor-saving, large screening capacity, direct access to antibody genes, ease of construction of various engineered antibodies, and expression in prokaryotic systems. In particular, it is of epoch-making significance to obtain antibodies against any antigen as well as human antibodies without immunization. The key to obtaining the expected specific antibodies is to build libraries that have large enough capacity and diversity and powerful screening tools. At present, the methods still need to be continuously improved and perfected. In this paper, we will give an overview of how to increase the capacity and diversity of libraries during the construction of antibody libraries and how to screen them.
