目的观察外源性IL-6对胆管癌细胞系QBC939凋亡及对Bcl-2 mRNA表达的影响。方法用MTT法检测外源性IL-6对胆管癌细胞QBC939的促增殖作用;通过annexin V/FITC和PI染色、流式细胞分析检测外源性IL-6对QBC939细胞凋亡的影响,RT-PCR测定Bcl-2 mRNA的表达。结果外源性IL-6能刺激QBC939细胞增殖,且QBC939细胞增殖与IL-6浓度呈正相关(r=1);外源性IL-6处理后,凋亡率显著低于对照组(P<0.05),外源性IL-6处理细胞后Bcl-2 mRNA显著高于对照组(P<0.05)。结论外源性IL-6在一定剂量范围内促进QBC939细胞生长,并抑制QBC939细胞凋亡;而此作用可能与上调Bcl-2 mRNA表达有关。
Objective To observe the effect of exogenous IL-6 on the apoptosis and the expression of Bcl-2 mRNA in cholangiocarcinoma cell line QBC939. Methods MTT assay was used to detect the effect of exogenous IL-6 on the proliferation of human cholangiocarcinoma QBC939 cells. Flow cytometry was used to detect the effect of exogenous IL-6 on the apoptosis of QBC939 cells by annexin V / FITC and PI staining. RT -PCR determination of Bcl-2 mRNA expression. Results Exogenous IL-6 could stimulate the proliferation of QBC939 cells, and the proliferation of QBC939 cells was positively correlated with the concentration of IL-6 (r = 1). The apoptosis rate of QBC939 cells after exogenous IL-6 treatment was significantly lower than that of the control group (P < 0.05). The expression of Bcl-2 mRNA in cells treated with exogenous IL-6 was significantly higher than that in control group (P <0.05). Conclusion Exogenous IL-6 can promote the growth of QBC939 cells and inhibit the apoptosis of QBC939 cells in a dose range, which may be related to the up-regulation of Bcl-2 mRNA expression.
