为了解老化过程中人类肺部基因表达谱改变 ,作者从老年和成年人正常肺组织提取总RNA ,用基因芯片技术比较基因表达差异。结果显示 ,研究的 8192条基因中有 87(1 0 6 % )条表达差异在 1 6倍以上 ,4 7(0 5 7% )条上调 ,4 0 (0 4 9% )条下调。上调为主的有 :细胞骨架和胞外基质、炎性应激反应、肿瘤相关、信号转导等 ;下调为主的有 :DNA/RNA合成修复、蛋白质合成修饰、能量代谢等 ;细胞周期与增殖相关基因上下调比例相当。提示老年人肺部基因表达谱发生了明显改变 ,与肺老化改变特征相符。
To understand the changes in gene expression profile of human lung during aging, the authors extracted total RNA from normal lung tissues of aged and adult adults and compared the gene expression differences using gene chip technology. The results showed that among the 8192 genes studied, 87 (106%) showed more than 16-fold difference, 47 (0 57%) were up-regulated and 40 (0 49%) were down-regulated. Up-based are: cytoskeleton and extracellular matrix, inflammatory stress response, tumor-related, signal transduction; down regulation are: DNA / RNA synthesis and repair, protein synthesis and modification, energy metabolism; cell cycle and Proliferation-related genes down a considerable proportion. It suggested that lung gene expression profiles of the elderly had a significant change, consistent with the characteristics of lung aging changes.
