[目的]探讨热休克蛋白90(HSP90)基因的表达与食管癌发生、发展和侵袭转移的关系及临床意义。[方法]通过Transwell侵袭小室分离得到侵袭能力更高的Eca-109T4,分别用Trizol提取mRNA,逆转录生产cDNA,用实时荧光定量PCR法检测HSP90mRNA的表达。[结果]通过Transwell侵袭小室筛选出高侵袭力的细胞亚株并命名为Eca-109T4。实时荧光定量PCR示HSP90在Eca-109T4升高(P<0.05)。[结论]HSP90mRAN表达水平,其在高转移的Eca-109T4上调,可能与食管癌的高转移有关,有望成为食管癌诊断与预后判断的指标。
[Objective] To investigate the relationship between the expression of heat shock protein 90 (HSP90) gene and the occurrence, development, invasion and metastasis of esophageal carcinoma and its clinical significance. [Method] Eca-109T4 with higher invasiveness was isolated by Transwell invasion chamber. MRNA was extracted by Trizol and cDNA was reverse transcribed. The expression of HSP90 mRNA was detected by real-time fluorescence quantitative PCR. [Result] The highly invasive cell sub-strain was screened by Transwell invasion chamber and named as Eca-109T4. Real-time fluorescence quantitative PCR showed HSP90 increased in Eca-109T4 (P <0.05). [Conclusion] The expression level of HSP90mRAN, which is up-regulated in highly metastatic Eca-109T4, may be related to the high metastasis of esophageal cancer and is expected to become an index of diagnosis and prognosis of esophageal cancer.
