小麦的加工品质与小麦高低分子量麦谷蛋白的组成和含量密切相关,特别是Glu-1位点编码的高分子量麦谷蛋白亚基(HMW-GS)的组成和含量。为此,试验利用1By8亚基标记对黄淮麦区目前生产利用或曾经利用过的小麦品种及部分育种资源共150份进行了检测,其中有43份材料扩增出527bp特异片段,表明具有1By8亚基;107份材料未扩增出527bp片段,表明不具有1By8亚基;1By8亚基的出现频率为28.6%。所用材料的鉴定结果与SDS-PAGE电泳的结果基本一致,表明利用特异性PCR标记鉴定1By8亚基是可靠的。与SDS-PAGE相比,PCR标记更加快速、简便和准确。
The processing quality of wheat is closely related to the composition and content of high and low molecular weight glutenin in wheat, especially the composition and content of high molecular weight glutenin subunit (HMW-GS) coded by Glu-1 locus. To this end, the test using 1By8 subunit markers of Huanghuai wheat currently produced or used in wheat varieties and parts of a total of 150 breeding resources were tested, of which 43 were amplified 527bp specific fragment, indicating that with 1By8 Subunit. The 527bp fragment was not amplified from 107 materials, indicating that there was no 1By8 subunit. The frequency of 1By8 subunit was 28.6%. The identification results of the materials used are basically the same as the results of SDS-PAGE electrophoresis, indicating that the identification of the 1By8 subunit by specific PCR markers is reliable. Compared with SDS-PAGE, PCR labeling is faster, easier and more accurate.
