杉木CAD片段为信息探针,利用序列拼接方法获得了738bp的cDNA序列.利用MEGA4.0软件对不同进化层次的代表植物相应CAD基因进行比对,构建系统进化树.利用生物信息学方法分析了杉木CAD基因的结构与功能,其中包括核苷酸序列的组成、ORF识别、CpG岛、限制性酶切位点分析、卷曲螺旋、亲水性、糖基位点及二级结构.结果表明:杉木与铁杉的CAD基因具有99.00%的同源性;该序列在88~737bp片段上有一个开放性阅读框,该序列编码的是一个碱性蛋白,亲水性较强,疏水性较弱,信号肽序列为第1位~第11位,二级结构以α-螺旋(27.50%)与不规则盘绕(41.50%)为主,延伸链(22.50%)也是该蛋白的重要结构元件,β转角区域仅为8.50%.
