应用MTT比色测定细胞活性的方法 ,观察药物对AGE形成的影响以及药物对AGE引起的牛视网膜微血管周细胞损伤的影响。结果葛根素及大黄素与BSA共孵后形成的糖基化产物对周细胞的损伤较不给药组有不同程度的减轻作用 ,终浓度为10mM效应最大 ,提示药物可能在孵育时抑制了AGE的形成 ;细胞培养基中直接加入葛根素 (1,10mM )能对AGE引起的周细胞损伤起一定的保护作用。
MTT colorimetric assay of cell activity was used to observe the effect of drugs on AGE formation and the effect of drugs on AGE-induced damage to bovine retinal microvascular pericytes. Results The glycosylation products formed after co-incubation of puerarin and emodin with BSA caused a lesser degree of damage to the pericytes than the untreated group. The final concentration was 10 mM, indicating that the drug may inhibit AGE during incubation. The formation of puerarin (1,10 mM) in cell culture medium can protect AGE-induced pericyte damage.
