目的 :研究冠状动脉支架术后高敏C反应蛋白 (hsCRP)的升高与树突状细胞 (DC)的关系。方法 :术前及术后 2d分离 11例冠状动脉支架术患者外周血单个核细胞 (PBMC) ,在含粒细胞巨噬细胞集落刺激因子(GM CSF)和白细胞介素 4 (IL 4 )的培养条件下制备DC。用流式细胞仪检测DC表面共刺激分子CD86 (B7 2 )的表达 ;混合淋巴细胞反应 (MLR)检测DC对同种异体T淋巴细胞的刺激能力 ;ELISA法测定MLR上清液中的细胞因子 ;比浊法测定hsCRP浓度 ,并探讨CD86表达与hsCRP的相关性。结果 :与术前比较 ,术后 2dDC表面CD86的表达明显增高 ;对T淋巴细胞刺激的能力增强 ;经DC刺激的淋巴细胞分泌促炎细胞因子 (IL 1β、IL 6、TNF α)增多 ,抑炎细胞因子 (IL 10 )减少 ;同时血hsCRP升高 ,且CD86的表达与血hsCRP水平正相关。结论 :支架术后DC的功能被激活 ,由此启动的T淋巴细胞的增殖和炎性反应可能是术后hsCRP升高的原因
Objective: To investigate the relationship between elevated hsCRP and dendritic cells (DC) after coronary stenting. Methods: Peripheral blood mononuclear cells (PBMCs) were isolated from 11 cases of coronary artery stenting before and 2 days after operation, and cultured in culture medium containing GM-CSF and IL-4 DC was prepared. The expression of costimulatory molecule CD86 (B7 2) on DCs was detected by flow cytometry. The stimulatory ability of DC on allogeneic T lymphocytes was detected by mixed lymphocyte reaction (MLR). The cytokines ; Turbidimetry hsCRP concentration, and to explore the CD86 expression and hsCRP correlation. Results: Compared with the preoperative level, the expression of CD86 on the surface of the postoperative 2 d DC was significantly increased, and the ability of stimulating T lymphocytes was enhanced. The number of proinflammatory cytokines (IL 1 β, IL 6 and TNF α) secreted by DCs stimulated by DC increased Inflammatory cytokines (IL 10) decreased; while blood hsCRP increased, and CD86 expression and serum hsCRP levels are positively correlated. CONCLUSIONS: The function of DC after activation of the scaffold is activated. The proliferation and inflammatory response of T lymphocytes activated by this technique may be the reason for the postoperative hsCRP increase
