目的:探讨右归饮对磨损颗粒诱导的大鼠假体周围骨溶解的影响。方法:选用40只成年雄性SD大鼠,随机分成4组,每组10只,分别为空白对照组、模型对照组、阿仑膦酸钠组和右归饮组。空白对照组不作任何处理;模型对照组行右侧股骨植入聚乙烯颗粒和钛棒制备聚乙烯颗粒诱导假体周围骨溶解大鼠模型;阿仑膦酸钠组每日空腹阿仑膦酸钠2 mL(蒸馏水与片剂的混悬液)灌胃,剂量为0.083mg/(kg·d),1次/d;右归饮治疗组同模型对照组,并以每100g大鼠灌胃2mL计算给药量,每周3次。8周后取血,然后处死所有实验动物以制备股骨标本,进行各组标本的TRAP染色和血清TRACP5b指标检测。结果:1)血清TRACP5b结果为右归饮和阿仑膦酸钠组可明显降低TRACP5b水平,与模型对照组比较差异有统计学意义(P<0.05)。2)TRAP染色阳性OC的观察和分析显示,光镜下右归饮和阿仑膦酸钠组TRAP阳性OC数均少于模型对照组,差异有统计学意义(P<0.05)。结论:右归饮能够有效抑制聚乙烯颗粒诱导的大鼠假体周围骨溶解。
Objective: To investigate the effect of Youguiyin on the osteolysis around rat prosthesis induced by wear particles. Methods: Forty adult male Sprague-Dawley rats were randomly divided into 4 groups (n = 10): blank control group, model control group, alendronate sodium group, and Yougui Yin group. Blank control group without any treatment; the model control group of right femur implanted polyethylene particles and titanium rods prepared polyethylene particles induced peri-implant osteolysis rats model; alendronate sodium group daily fasting alendronate sodium 2 mL (distilled water and tablet suspension) gavage, a dose of 0.083mg / (kg · d), 1 / d; Youguiyin treatment group with the model control group, and every 100g rats were intragastric 2mL Calculate the dose, 3 times a week. After 8 weeks, the blood was taken out, and then all experimental animals were sacrificed to prepare femoral specimens. TRAP staining and TRACP5b detection of serum were performed in each group. Results: 1) The results of TRACP5b in serum were significantly lower than those in the control group (P <0.05). 2) The observation and analysis of TRAP-positive OC showed that the number of TRAP-positive OC in both right-handed and alendronate groups was less than that in model control group under light microscope (P <0.05). Conclusion: Yougui Yin can effectively inhibit the polyethylene particles induced osteolysis around the rat prosthesis.
