目的探讨肾性高血压模型大鼠(RHR)血浆前列腺素(PGI2)、血栓素(TXA2)水平的变化及首乌降压胶囊的降压作用机制。方法采用二肾一夹(2K1C)法复制RHR模型,观察血浆6-Keto-PGF1α、TXB2水平。结果首乌降压胶囊高、低剂量组和卡托普剂组治疗后血压均明显低于模型组,有显著差异(P<0.001,P<0.05,P<0.001),说明首乌降压胶囊和卡托普利均有较好的降压效果;与模型组比较,首乌降压胶囊高、低剂量组均使RHR血浆6-Keto-PGF1α水平显著升高(P<0.01,P<0.05),而TXB2水平显著下降(P<0.01,P<0.05)。结论RHR高血压形成与血浆6-Keto-PGF1α水平下降、TXB2水平升高有关,首乌降压胶囊能有效降低血压,其降压机制可能与调整血浆PGI2、TXA2水平有关。
Objective To investigate the changes of plasma prostaglandin (PGI2) and thromboxane (TXA2) levels and the antihypertensive mechanism of Shouwu Jiangya capsule in rats with renal hypertension (RHR). Methods Two-kidney and one clip (2K1C) method was used to replicate the RHR model. The levels of plasma 6-Keto-PGF1α and TXB2 were observed. Results The blood pressure of Shouwu Jiangya capsule high and low dose group and captopril group was significantly lower than that of the model group after treatment (P<0.001, P<0.05, P<0.001), indicating that Shouwu Jiangya capsules Both captopril and carbamazepril had good antihypertensive effect. Compared with the model group, the levels of 6-Keto-PGF1α in plasma of RHR were significantly increased in the high and low dose groups of Shouwu Jiangya capsule (P<0.01, P<0.05). ), and TXB2 levels decreased significantly (P <0.01, P <0.05). Conclusion The formation of hypertension in RHR is related to the decrease of plasma 6-Keto-PGF1α level and TXB2 level. Shouwu Jiangya Capsule can effectively reduce blood pressure, and its antihypertensive mechanism may be related to the adjustment of plasma PGI2 and TXA2 levels.
