目的研究白细胞介素-18(IL-18)在大鼠重症急性胰腺炎(SAP)中的表达。探讨其血清水平与胰腺组织损伤程度的相关性。方法采用胰腺被膜下注射牛磺胆酸钠(Na-Tc)建立急性胰腺炎模型。60只SPF级雄性W istar大鼠随机分为空白组、假手术组、胰腺炎6 h、12 h及24 h 5组。动态测定血淀粉酶、血清IL-18及胰腺组织病理评分。结果假手术组与空白组比较差异无统计学意义(P>0.05);胰腺炎6 h、12 h、24 h组与空白组比较,差异均有统计学意义(P<0.01),且组间两两比较差异有统计学意义(P<0.05)。血清IL-18含量与胰腺组织病理评分有高度相关(r>0.7)。结论大鼠重症急性胰腺炎组血清淀粉酶增高明显,与胰腺组织病理学改变相一致;IL-18在血清中含量与胰腺病理评分存在正相关。IL-18在SAP病理损伤中起重要作用。
Objective To study the expression of interleukin-18 (IL-18) in severe acute pancreatitis (SAP) in rats. To investigate the correlation between serum levels and the degree of pancreatic tissue damage. Methods Pancreatic subconjunctival injection of sodium taurocholate (Na-Tc) was used to establish acute pancreatitis model. Sixty SPF male Wistar rats were randomly divided into blank group, sham operation group, pancreatitis 6 h, 12 h and 24 h 5 groups. Dynamic determination of serum amylase, serum IL-18 and pancreatic histopathological score. Results There was no significant difference between the sham group and the blank group (P> 0.05). There was significant difference between the sham operation group and the blank group at 6 h, 12 h and 24 h (P <0.01) The difference was statistically significant (P <0.05). Serum IL-18 levels and pancreatic histopathological scores were highly correlated (r> 0.7). Conclusion Serum amylase increased significantly in severe acute pancreatitis group, which is consistent with the histopathological changes of pancreas. The content of IL-18 in serum is positively correlated with the pathological score of pancreatic tissue. IL-18 plays an important role in SAP pathological injury.
