本实验以高秆野生稻的幼嫩叶鞘和茎为材料。外植体在N_6添加NAA0.5mg/l、2,4-D2mg/l、蔗糖3%的培养基上诱导愈伤组织。20至25天以后,把愈伤组织转入两种MS培养基诱导分化,一种添加NAA2mg/l、IAA0.5mg/l、蔗糖6%;另一种添加NAA2mg/l、IAA0.5 mg/l、KT2mg/l、蔗糖6%。叶鞘和茎愈伤组织都能在这两种培养基上分化出完整的植株。出愈率与外植体的器官来源有关。愈伤组织类型是影响植株分化的重要因素。愈伤组织的器官来源和培养基也对植株分化有影响。
In this experiment, young leaf sheaths and stems of high-stalk wild rice were used as materials. The explants were induced to callus on N6 medium supplemented with NAA0.5mg / l, 2,4-D2mg / l and sucrose3%. After 20 to 25 days, the callus was transferred to two MS mediums to induce differentiation, one with NAA 2 mg / l, IAA 0.5 mg / l and sucrose 6%, and the other with NAA 2 mg / l and IAA 0.5 mg / l, KT2mg / l, sucrose 6%. Both sheath and stem callus differentiated intact on both media. The healing rate is related to the organ origin of the explant. Callus type is an important factor affecting plant differentiation. The organ sources and media for callus also have an effect on plant differentiation.
