目的 探讨分化诱导剂六亚甲基二乙酰胺 (HMBA)诱导人粘液表皮样癌细胞后 ,p2 1、增殖细胞核抗原 (PCNA)和CyclinD1 表达的变化。方法 用 2mmol/LHMBA对培养的粘液表皮样癌细胞进行体外诱导 72h后 ,分别采用免疫组织化学、图像分析等方法对处理与未处理细胞的p2 1、PCNA和CyclinD1 表达变化进行定量分析。结果 HMBA处理的粘液表皮样癌细胞p2 1表达明显高于对照组 (P <0 .0 1 ) ;PCNA和CyclinD1 表达明显低于对照组(P <0 .0 1 )。结论 HMBA通过影响p2 1、PCNA和CyclinD1 的表达而发挥对粘液表皮样癌细胞的抑制增殖作用
Objective To investigate the expression of p21, proliferating cell nuclear antigen (PCNA) and CyclinD1 in human mucoepidermoid carcinoma cells induced by differentiation induced by hexamethylene bisacetamide (HMBA). Methods The mucoepidermoid carcinoma cells were induced with 2mmol / LHMBA for 72 hours in vitro. The expressions of p21, PCNA and CyclinD1 were analyzed by immunohistochemistry and image analysis respectively. Results The expression of p21 in HMBA-treated mucoepidermoid carcinoma cells was significantly higher than that in the control group (P <0.01). The expressions of PCNA and CyclinD1 in HMBA-treated mucoepidermoid carcinoma cells were significantly lower than those in the control group (P <0.01). Conclusion HMBA can inhibit the proliferation of mucoepidermoid carcinoma cells by affecting the expression of p21, PCNA and CyclinD1
