目的研究灵芝多糖在体外对人类白血病细胞株NB4细胞的增长抑制及凋亡的影响及抗肿瘤的机制。方法用MTT比色法检测灵芝多糖对NB4细胞增长的抑制作用,Annexin V-FITC/PI双染色法检测NB4细胞的凋亡情况,荧光定量PCR及蛋白印迹技术检测早期生长反应因子1(Egr-1)基因的表达。结果灵芝多糖对NB4细胞增殖具有抑制作用并可促其凋亡,最高分别为33.5%及18.77%。这种效应呈时间-剂量依赖性。与对照组比较,差异有统计学意义(P<0.01)。对Egr-1 mRNA和蛋白表达也具有明显的促进作用,差异有统计学意义(P<0.01)。结论灵芝多糖在体外能够明抑制NB4细胞的增长并促其凋亡,其机制可能与上调Egr-1的表达相关。
Objective To study the effects of Ganoderma lucidum polysaccharides on the growth inhibition and apoptosis of human leukemia cell line NB4 in vitro and the anti-tumor mechanism. Methods The inhibitory effect of Ganoderma lucidum polysaccharides on the growth of NB4 cells was detected by MTT colorimetric assay. The apoptosis of NB4 cells was detected by Annexin V-FITC / PI double staining. The expression of Egr- 1) Gene expression. Results Ganoderma lucidum polysaccharide could inhibit the proliferation of NB4 cells and promote the apoptosis of NB4 cells with the highest levels of 33.5% and 18.77%, respectively. This effect is time-dose dependent. Compared with the control group, the difference was statistically significant (P <0.01). Egr-1 mRNA and protein expression also had a significant role in promoting, the difference was statistically significant (P <0.01). Conclusion Ganoderma lucidum polysaccharide can inhibit the growth of NB4 cells in vitro and promote its apoptosis, which may be related to the up-regulation of Egr-1 expression.
