目的:探讨黄芪注射液对重症肺炎大鼠胸腺细胞凋亡相关蛋白表达的影响。方法:将40只Sprague-Dawley(SD)大鼠随机分为对照组、模型组、黄芪高剂量组、黄芪中剂量组和黄芪低剂量组,每组8只。采用气管穿刺注入感染法制作大鼠重症肺炎模型。分别采用原位末端转移酶标记法(Terminal deoxynucleotidyl transfer-ase dUTP nick end labeling,TUNEL)检测各组大鼠胸腺细胞凋亡及Western blot检测凋亡相关蛋白cleaved caspase-3、Bcl-2和Bax的表达。结果:与对照组比较,模型组和黄芪治疗各组胸腺细胞凋亡增加,凋亡相关蛋白cleaved caspase-3和Bax表达增强,除黄芪高剂量组外Bcl-2表达均减弱,Bcl-2/Bax比值降低(P<0.05);与模型组比较,黄芪高剂量组胸腺细胞凋亡减轻,Bcl-2表达增强,cleaved caspase-3和Bax表达减弱(P<0.05),Bcl-2/Bax比值增加。结论:黄芪注射液能减轻重症肺炎时胸腺细胞凋亡,其机制与促进Bcl-2表达,减轻Bax表达,增加Bcl-2/Bax比值有关。
Objective: To investigate the effect of Astragalus injection on thymocyte apoptosis-related protein expression in severe pneumonia rats. Methods: Forty Sprague-Dawley (SD) rats were randomly divided into control group, model group, Astragalus high dose group, Astragalus middle dose group and Astragalus low dose group, with 8 rats in each group. Rat model of severe pneumonia was made by tracheal injection and infection. The apoptosis of thymocytes in each group was detected by TUNEL, and the expressions of cleaved caspase-3, Bcl-2 and Bax were detected by Western blot expression. Results: Compared with the control group, the apoptosis of thymus cells and the expression of cleaved caspase-3 and Bax in model group and Astragalus membranaceus group were enhanced, while the expression of Bcl-2 / (P <0.05). Compared with the model group, the apoptosis of thymus in high dose Astragalus group was alleviated, the expression of Bcl-2 was increased, the expression of cleaved caspase-3 and Bax was weakened, and the ratio of Bcl-2 / increase. Conclusion: Astragalus injection can reduce the apoptosis of thymocytes in severe pneumonia. The mechanism may be related to promoting the expression of Bcl-2, decreasing the expression of Bax and increasing the ratio of Bcl-2 / Bax.
