目的:分析中国南方食管癌高发地区(潮汕地区)食管鳞状细胞癌APC基因连锁位点D5S346和D5S82的杂合性丢失情况。方法:采用PCR银染技术,检测42例配对食管鳞癌标本D5S346和D5S82的杂合性丢失情况。结果:D5S346和D5S82在该食管癌人群中杂合性丢失率高达35%和39%。结论:APC基因附近微卫星标记D5S346和D5S82在中国南方食管鳞癌中的杂合性丢失频率较高,提示APC基因在食管癌中可能同样存在缺失改变。
OBJECTIVE: To analyze the loss of heterozygosity at the linkage sites of APC gene D5S346 and D5S82 in esophageal squamous cell carcinoma (ESCC) with high prevalence in esophageal cancer in southern China. Methods: The loss of heterozygosity of 42 matched esophageal squamous cell carcinoma specimens D5S346 and D5S82 was detected by PCR silver staining. Results: The loss of heterozygosity of D5S346 and D5S82 in this population of esophageal cancer was as high as 35% and 39%, respectively. Conclusion: The loss of heterozygosity of microsatellite markers D5S346 and D5S82 near the APC gene in esophageal squamous cell carcinoma in southern China is high, suggesting that there may be a deletion change of APC gene in esophageal cancer.
