探讨了乳酸克鲁维酵母(K.Lactic)β- 半乳糖苷酶金属离子稳定性和热稳定性.证明某些金属离子对β-半乳糖苷酶具激活作用,另一些金属离子对β-半乳糖苷酶活性影响不大,但是在高浓度的条件下均表现出对酶活性不同程度的抑制作用.Ca2+ 对酶的抑制可以被磷酸盐、脱脂乳以及Mg2+ ,Mn2+ 所恢复.K+ 单独存在就可以大大地提高酶的热稳定性,Mn2+ 可以增加K+ 对β-半乳糖苷酶的热稳定性作用,而Mg2+ ,Mn2+ 和Na+ 自身不能提高酶的热稳定性.甘油、脱脂乳均可以提高酶的热稳定性,脱脂乳成分如乳糖、酪蛋白酸钠对酶的热稳定作用有赖于K+ 的存在
The metal ion stability and thermostability of K. lactis β-galactosidase were discussed. Some metal ions proved that β-galactosidase had an activation effect, while other metal ions had little effect on the β-galactosidase activity, but exhibited inhibitory effects on the enzyme activities to a great extent under high-concentration conditions . The inhibition of Ca2 + on the enzyme can be restored by phosphates, skimmed milk, and Mg2 +, Mn2 +. The presence of K + can greatly improve the thermostability of the enzyme. Mn2 + can increase the thermostability of β-galactosidase by K +, while Mg2 +, Mn2 + and Na + do not improve the thermostability of the enzyme. Glycerol, skim milk can improve the thermal stability of the enzyme, skim milk components such as lactose, sodium caseinate thermal stability of the enzyme depends on the existence of K +
