丹参对大鼠局灶性脑缺血后神经元内细胞周期蛋白D1表达的影响

目的 观察大鼠局灶性脑缺血后神经元内细胞周期蛋白D1(cyclinD1)表达与神经细胞凋亡的关系以及丹参的影响。方法 采用线栓法大鼠大脑中动脉持续阻塞模型 ,应用免疫组织化学和TUNEL染色方法观察假手术组和缺血后不同时间阳性染色神经细胞的数量和分布情况及丹参的影响。结果 cyclinD1蛋白表达自缺血后 6h开始出现 ,缺血后 48h达高峰 ;丹参组各相应时间点cyclinD1蛋白表达明显减少 (P <0 .0 1)。凋亡细胞自缺血后 12h开始出现 ,缺血后 72h达高峰 ;丹参组各相应时间点凋亡细胞表达明显减少 (P <0 .0 1)。相邻切片可见cyclinD1蛋白表达和凋亡细胞染色区域基本相同。结论 脑缺血后cyclinD1的异常表达可能诱发了缺血神经细胞的凋亡 ,丹参可能通过抑制cyclinD1的表达而减少缺血后神经细胞的损害 Objective To observe the relationship between the expression of cyclin D1 in neurons and the apoptosis of neurons in rats after focal cerebral ischemia and the effect of Salvia miltiorrhiza. Methods The middle cerebral artery occlusion model was established by thread occlusion. The number and distribution of positive staining neurons in sham operation group and different time after ischemia were observed by immunohistochemistry and TUNEL staining and the effect of Salvia miltiorrhiza. Results The expression of cyclinD1 protein began to appear at 6h after ischemia and peaked at 48h after ischemia. The expression of cyclinD1 in Salvia miltiorrhiza group was significantly decreased at the corresponding time points (P <0.01). Apoptotic cells began to appear at 12h after ischemia and peaked at 72h after ischemia. The apoptotic cells in Salvia miltiorrhiza group were significantly decreased at the corresponding time point (P <0.01). The adjacent sections showed that the expression of cyclinD1 protein was similar to that of apoptotic cells. Conclusion The abnormal expression of cyclinD1 may induce the apoptosis of ischemic neurons after cerebral ischemia. Salvia miltiorrhiza may reduce the damage of ischemic neuronal cells by inhibiting the expression of cyclinD1
