目的探讨程氏蠲痹汤加减方含药血清对体外培养的佐剂性关节炎(adjuvant arthritis,AA)大鼠外周血单核细胞中肿瘤坏死因子-α(tumor necrosis factor alpha,TNF-α)和白细胞介素-1β(interleukin-1beta,IL-1β)表达的影响。方法将清洁级雄性SD大鼠随机分成6组:胎牛血清组、程氏蠲痹汤加减方(JBT)组、雷公藤多苷片(TGT)组、JBT+sc-13143组、正常大鼠血清组以及阴性对照组。采用皮内注射弗氏完全佐剂和冷水吹风刺激的方法复制风寒湿痹阻型AA模型。检测各组大鼠血清培养基中外周血单核细胞TNF-α和IL-1β的表达水平。结果 JBT组大鼠外周血单核细胞TNF-α和IL-1β表达水平明显降低,与胎牛血清组、正常大鼠血清组以及阴性对照组比较,差异均有统计学意义(P<0.05)。JBT+sc-13143组与JBT组TNF-α、IL-1β表达水平相比,差异也有统计学意义(P<0.05)。结论程氏蠲痹汤加减方能有效下调AA大鼠外周血单核细胞TNF-α和IL-1β水平。
Objective To investigate the effect of Chengshi Huibi Decoction (MDC) containing serum on the expression of tumor necrosis factor alpha (TNF-α) in peripheral blood mononuclear cells of adjuvant arthritis (AA) And interleukin-1β (IL-1β) expression. Methods Male Sprague-Dawley rats were randomly divided into 6 groups: fetal bovine serum group, JBT group, TGT group, JBT + sc-13143 group, normal rats Serum group and negative control group. A cold rheumatoid arthritis model was replicated by intradermal injection of Freund’s complete adjuvant and cold water stimulation. The levels of TNF-α and IL-1β in peripheral blood mononuclear cells were detected in serum of each group. Results The levels of TNF-α and IL-1β in peripheral blood mononuclear cells of JBT group were significantly lower than those of the normal control group and the fetal bovine serum group (P <0.05) . The difference of TNF-α and IL-1β expression between JBT + sc-13143 group and JBT group was also statistically significant (P <0.05). Conclusion Chengbianbibi Decoction can effectively reduce the level of TNF-α and IL-1β in peripheral blood mononuclear cells of AA rats.
