目的 观察联合应用反义 bcl- 2硫代磷酸寡脱氧核苷酸 (bcl- 2 AS- PS- ODN)和白细胞介素 6 (IL - 6 )AS- PS- ODN对小细胞肺癌细胞株 NCI- H4 4 6增殖和活力的影响。 方法 以 bcl- 2 AS- PS- ODN和 IL- 6 AS- PS-ODN单独及联合作用于 NCI- H4 4 6 ,经细胞克隆形成实验、细胞增殖实验观察其对细胞增殖和活力的影响 ,经细胞凋亡线粒体流式检测试剂盒检测细胞凋亡的变化 ,经半定量 RT- PCR检测 IL - 6 m RNA表达水平的变化。 结果 细胞增殖曲线、细胞克隆形成实验及细胞凋亡实验显示联合用药能更强地抑制细胞活力 ,促进细胞凋亡。 bcl- 2 AS-PS- ODN单独作用时 ,IL- 6 m RNA表达上升 74 .3% ,IL- 6 AS- PS- ODN单独作用以及与 bcl- 2 AS- PS- ODN联合作用时 ,IL- 6分别下调 6 7.7%和 5 2 .4 %。 结论 联合用药较单独用药对 NCI- H4 4 6细胞株显示出更好的抑制效果
Objective To observe the effect of combined application of antisense bcl-2 phosphorothioate oligodeoxynucleotides (bcl-2 AS-PS-ODN) and interleukin-6 (AS-PS- ODN) on small cell lung cancer cell lines NCI- H4 4 6 Effects of proliferation and viability. Methods The effects of bcl-2 AS-PS-ODN and IL-6 AS-PS-ODN alone and in combination on NCI-H4 4 6 cells were observed by cell clone formation assay and cell proliferation assay. Apoptosis mitochondrial flow cytometry kit was used to detect the change of apoptosis. The expression of IL - 6 mRNA was detected by semi - quantitative RT - PCR. Results The cell proliferation curve, cell clone formation assay and apoptosis assay showed that combination therapy could inhibit cell viability and promote apoptosis. When bcl-2 AS-PS-ODN was administered alone, IL-6 mRNA expression increased by 74.3%, IL-6 AS-PS-ODN alone and combined with bcl- 6 down by 6 7.7% and 5.24% respectively. Conclusion Compared with the single drug alone, the combined drugs have better inhibitory effect on NCI-H4 4 6 cell lines
