目的:探讨补肾壮骨中药对模拟失重条件下体外培养成骨细胞的影响,拟在细胞水平上揭示该方对抗骨丢失的机制。方法:分离培养乳鼠颅骨成骨细胞,置回旋器30r/min,连续回旋模拟失重,体外含药血清干预3d,观察该方对失重成骨细胞碱性磷酸酶(ALP)和骨钙素(BGP)生成及细胞凋亡的影响。结果:与正常对照组比较,失重模型组成骨细胞在回旋3d细胞培养液中的ALP活性和BGP活性均显著降低(P<0.05);成骨细胞凋亡率显著增加(P<0.05)。与失重模型组比较,中药含药血清组ALP活性和BGP活性均明显升高,其中含药血清20%浓度组差异显著(P<0.05);成骨细胞凋亡率均减少,含药血清10%和20%浓度组差异显著(P<0.05)。结论:该方能改善模拟失重条件下成骨细胞的功能,减少成骨细胞凋亡,缓解其分化抑制状态,促进骨基质的形成和成熟。
OBJECTIVE: To investigate the effect of Bushenzhuanggu Chinese medicine on osteoblast cultured in vitro under simulated weightlessness conditions, and to reveal the mechanism of its side against bone loss at the cellular level. METHODS: Rat calvarial osteoblasts were isolated and cultured, and placed in a gyrator for 30 r/min. The weightlessness was simulated by successive convolutions. The in vitro serum containing drugs was intervened for 3 days to observe the effect of this side on alkaline phosphatase (ALP) and osteocalcin in weightless osteoblasts. BGP) production and apoptosis. RESULTS: Compared with the normal control group, the ALP activity and BGP activity in the 3d cell culture medium were significantly decreased (P<0.05); the osteoblast apoptosis rate was significantly increased (P<0.05). Compared with the weightlessness model group, the ALP activity and BGP activity in the TCM serum-containing serum group were significantly increased, and the concentration of the serum-containing serum 20% group was significantly different (P<0.05); the osteoblast apoptosis rate was reduced, and the drug-containing serum was reduced. There was a significant difference between the % and 20% concentrations (P<0.05). Conclusion: This method can improve the function of osteoblast under simulated weightlessness condition, reduce the osteoblast apoptosis, relieve the differentiation inhibition state, and promote the formation and maturation of bone matrix.
