目的:探讨白藜芦醇对人口腔鳞癌KB细胞的增殖及凋亡影响。方法:应用MTT比色法检测Res对KB细胞体外生长的抑制作用;流式细胞仪测定细胞的周期和凋亡;Hoechst33258荧光染料染色,显示凋亡细胞形态;Western blot方法检测survivin、Caspase-3、Smac蛋白的表达。结果:Res能以剂量和时间依赖的方式抑制KB细胞生长,以剂量依赖的方式使KB细胞周期阻滞在S期、诱导细胞凋亡;Hoechst33258染色,可见细胞呈明显的凋亡特征;Western blot方法测得Res下调KB细胞的Survivin蛋白表达水平、增加KB细胞胞浆的Smac蛋白含量(P<0.01)、同时活化Caspase-3蛋白。结论:Res可抑制KB增殖,使细胞周期呈S阻滞并可诱导细胞发生凋亡,Survivin、Caspase-3和Smac基因参与了Res诱导KB细胞凋亡的作用。
Objective: To investigate the effects of resveratrol on the proliferation and apoptosis of human oral squamous cell carcinoma KB cells. Methods: MTT assay was used to detect the inhibitory effect of Res on the growth of KB cells in vitro. The cell cycle and apoptosis were detected by flow cytometry. The morphological changes of apoptotic cells were observed by Hoechst33258 fluorescent dye staining. The expressions of survivin, Caspase-3 , Smac protein expression. Results: Res inhibited the growth of KB cells in a dose-and time-dependent manner, and blocked the cell cycle of KB cells in a dose-dependent manner to induce apoptosis. Hoechst33258 staining showed obvious apoptotic characteristics of KB cells. Western blot The results showed that Res down-regulated the expression of Survivin in KB cells and increased the content of Smac in cytoplasm of KB cells (P <0.01), and activated Caspase-3 protein. Conclusion: Res can inhibit the proliferation of KB, block the cell cycle and induce cell apoptosis. Survivin, Caspase-3 and Smac genes are involved in Res-induced KB cell apoptosis.
