目的研究邻苯二甲酸二乙基己酯(Di-2-ethylhexyl phthalate,DEHP)和双酚A(Bisphenol A,BPA)联合暴露对大鼠肝脏的影响。方法将Wistar大鼠随机分4组,分别灌胃1 500mg/kg DEHP、200mg/kg BPA及750mg/kg DEHP+100mg/kg BPA染毒6周。染毒终点测定血清中碱性磷酸酶(ALP)、胆固醇(CHO)、甘油三酯(TG)及镁离子(Mg2+)含量;检测肝匀浆中谷胱甘肽(GSH)、丙二醛(MDA)含量。结果与对照组比较,DEHP明显升高血清ALP、CHO及Mg2+含量,而BPA明显增加肝中MDA含量(均P<0.05)。与对照组比较,DEHP+BPA联合染毒除明显增加肝脏脏器系数外,对大鼠体重、肝MDA、血脂及血清Mg2+和ALP含量均无明显影响。与DEHP+BPA联合染毒组比较,BPA显著升高肝脏MDA含量;DEHP增加血清Mg2+含量差异均有统计学意义(均P<0.05)。结论 DEHP和BPA单独染毒均可产生肝损伤,其联合暴露产生交互作用。
Objective To investigate the effects of combined exposure of Di-2-ethylhexyl phthalate (DEHP) and bisphenol A (BPA) on rat liver. Methods Wistar rats were randomly divided into 4 groups and were administered with 500 mg / kg DEHP, 200 mg / kg BPA and 750 mg / kg DEHP + 100 mg / kg BPA respectively for 6 weeks. Serum levels of alkaline phosphatase (ALP), cholesterol (CHO), triglycerides (TG) and magnesium ions (Mg2 +) were measured at the end of treatment. Glutathione (GSH), malondialdehyde )content. Results Compared with the control group, DEHP significantly increased serum ALP, CHO and Mg2 + content, while BPA significantly increased the content of MDA in the liver (all P <0.05). Compared with the control group, addition of DEHP + BPA combined with the addition of significantly increased liver organ coefficient, the body weight, liver MDA, blood lipid and serum Mg2 + and ALP content had no significant effect. Compared with DEHP + BPA group, BPA significantly increased liver MDA content; DEHP increased serum Mg2 + content differences were statistically significant (P <0.05). Conclusions Both DEHP and BPA can induce liver injury by exposure to a single agent, and their combined exposure may produce an interaction.
