目的观察早期应用前列腺素E_1(PGE_1)对家兔单侧后肢电烧伤后局部股静脉血中的红细胞变形性、血小板聚集率和血管内皮素-1以及损伤肌肉组织中的ATP含量变化的影响。方法采用家兔单侧后肢电烧伤模型,分别用激光衍射法、放射兔疫法和高压液相色谱等方法对实验指标进行测定。结果肢体电烧伤后,局部深静脉血中的红细胞变形性(RCD)、血小板聚集率(PAR)和血管内皮素-1(ET-1)的含量均有明显改变。早期应用PGE_1可显著改善电烧伤组织深静脉血中的RCD,降低PAR以及ET-1的含量,并提高损伤肌肉组织中的ATP含量,其与未治疗组及对侧健肢相比均有非常显著差别。结论肢体高压电烧伤后,局部血液中有明显的血液流变学变化,早期应用PGE_1可减轻电烧伤组织中血液的超凝血状态,预防大血管继发血栓形成,保护间生态组织。
Objective To observe the effects of early application of prostaglandin E_1 (PGE_1) on erythrocyte deformability, platelet aggregation rate, endothelin - 1 and ATP content in damaged muscle tissue in local femoral vein after unilateral hindlimb electric burn in rabbits. Methods The model of unilateral hindlimb electric burn in rabbits was used to determine the experimental parameters by laser diffraction, radioimmunoassay and high pressure liquid chromatography. Results After limb burns, the RCD, PAR and ET-1 in deep venous blood changed significantly. Early application of PGE_1 significantly improved RCD, decreased PAR and ET-1 levels in deep venous blood of electrically-burned tissue, and increased ATP content in injured muscle tissue compared with untreated and contralateral limbs Significant difference. Conclusion The limbs have obvious hemorheological changes in the local blood after the high voltage electric burns on the limbs. Early use of PGE_1 can reduce the hypercoagulability state of the blood in the electric burn tissue, prevent the secondary thrombosis of large vessels and protect the ecological tissues.
