在对体外新生大白鼠(0~1d)大脑皮层神经细胞进行原代培养的基础上,分别加入不同浓度苯巴比妥(PB),继续培养3d。观察活细胞形态、计数活细胞及检测细胞DNA,RNA含量。结果显示:高浓度PB(≥60mg/L)对神经细胞生长具有明显抑制作用(P<0.01),高浓度PB(120mg/L)可引起细胞DNA,RNA含量明显减低(P<0.01)。提示高浓度PB对神经细胞存在有毒性作用。
On the basis of the primary culture of neurons in the neonate rat (0 ~ 1d) in vitro, different concentrations of phenobarbital (PB) were added separately and cultured for 3 days. Live cell morphology was observed. Counting of living cells and detection of DNA and RNA content. The results showed that high concentration of PB (≥60mg / L) significantly inhibited the growth of neurons (P <0.01). High concentrations of PB (120mg / L) could significantly reduce the DNA and RNA content of cells (P <0.01). Tip high concentrations of PB on the existence of toxic effects of nerve cells.
