核酸酶P1是一种重要的DNA与RNA水解酶。本文利用ICP、VIS、NMR、酶活性测定等分析方法,首次拓展研究了核酸酶P1与CoCl2的直接相互作用。结果发现:核酸酶P1活性中心Zn?离子可被外加CoCl2中的Co?部分取代,而Co?进入酶的活性中心,生成相应的酶衍生物“Co?-P1”,并影响了酶的催化活性。与此同时还获得了Co?进入核酸酶P1活性中心Zn2位上的酶衍生物1HNMR谱。
Nuclease P1 is an important DNA and RNA hydrolase. In this paper, the direct interaction between nuclease P1 and CoCl2 was first explored using ICP, VIS, NMR and enzyme activity assays. The results showed that the Zn? Ion of the active site of nuclease P1 could be replaced by the Co? Moiety of CoCl2 and Co? Entered the active site of the enzyme to generate the corresponding enzyme derivative & quot; Co? -P1 & active. In the meantime, the 1H NMR spectrum of the enzyme derivative entering the Zn2 position of the active site of nuclease P1 was also obtained.
