目的 :探讨诱导分化剂全反式维甲酸 (ATRA)联合α -干扰素 (α -IFN )对人涎腺腺样囊性癌细胞的作用效能 ,寻找治疗腺样囊性癌的新途径。方法 :应用ATRA和α -IFN ,分别以单用、两两联用对腺样囊性癌SACC -83细胞株作用 ,用MTT法、软琼脂集落形成、BrdU掺入法和图像分析的方法观察细胞的形态和功能变化。结果 :①两药联用时较两药分别单用时对腺样囊性癌细胞的抑制率升高 ,平板克隆形成率下降 ,DNA合成受抑制。②癌细胞恶性表型逐渐消失 ,成熟细胞特征逐渐增多。③药物作用后细胞凋亡现象明显 ,可见大量凋亡小体。结论 :两种药物联合使用时可对腺样囊性癌细胞产生显著的抑制增殖、诱导分化和促凋亡作用。全反式维甲酸联合α -干扰素对腺样囊性癌细胞有明显的抗肿瘤作用。
Objective: To explore the effect of inducing differentiation of ATRA and α - IFN on human salivary adenoid cystic carcinoma cells and to find a new way to treat adenoid cystic carcinoma. Methods: ATRA and α -IFN were used alone or in combination with adenosine cystic carcinoma cell line SACC-83 respectively. MTT assay, soft agar colony formation assay, BrdU incorporation assay and image analysis Cell morphology and function changes. Results: ① When combined with two drugs, the inhibitory rate of adenoid cystic carcinoma cells increased, the rate of plate clone formation decreased and DNA synthesis was inhibited. ② malignant phenotype of cancer cells gradually disappear, the characteristics of mature cells gradually increased. ③ After the drug effect of apoptosis was obvious, we can see a large number of apoptotic bodies. Conclusion: The combination of two drugs can significantly inhibit proliferation, induce differentiation and promote apoptosis of adenoid cystic carcinoma cells. All - trans retinoic acid combined with α - interferon has obvious anti - tumor effect on adenoid cystic carcinoma cells.
