以酵母整合型质粒YIP5-Km为载体,从HindⅢ消化的水稻(珍山97不育系)DNA中分离到一些在酵母细胞中能自主复制的DNA片段(Autonomously Replication Sequence,简称ARS).其中ARS2为4.0 kb,ARS7为14.2 kb. 用Southern杂交证明它们来源于水稻的核DNA,而且它们在水稻核DNA中是高度重复序列.文中还对ARS2和ARS7进行了亚克隆分析,结果表明ARS2的三个亚克隆片段ARS2-1,ARS2-2,ARS2-3都是高度重复序列,而且ARS2-2,ARS2-3还具有ARS功能;ARS7的三个亚克隆片段中只有ARS7-1是高度重复序列但无ARS功能,ARS7-3有ARS功能但不是高度重复序列.
Autonomousously Replication Sequence (ARS) was isolated from DNA of HindIII-digested rice (Zhenshan 97 male sterile line) with yeast-integrating plasmid YIP5-Km as a vector, and ARS2 Was 4.0 kb and ARS7 was 14.2 kb. Southern hybridization proved that they originated from nuclear DNA in rice and that they were highly repetitive sequences in rice nuclear DNA.The subclones of ARS2 and ARS7 were subcloned and the results showed that ARS2 Each of the subclone fragments ARS2-1, ARS2-2 and ARS2-3 are highly repetitive sequences, and ARS2-2 and ARS2-3 also have ARS functions.
