目的观察芪薏益肝煎对H22荷瘤小鼠的抑瘤作用,并探讨其作用机制。方法小鼠右腋下皮下种瘤造成实体瘤模型,将30只小鼠分为3组,对照组、中药组和5-Fu组各10只。用药15 d后取瘤称重计算抑瘤率、胸腺指数、脾指数;采用免疫组化技术检测CyclinD1和Caspase-3的表达。结果芪薏益肝煎对小鼠肝癌细胞的生长抑制率为37.28%,5-Fu组为50.48%。Cyclin D1及Caspase-3在3组中均呈阳性表达,统计结果表明,中药组CyclinD1表达减弱,Caspase-3表达增强,与对照组比较差异有统计学意义。结论芪薏益肝煎对肝癌细胞的增殖有明显的抑制作用,其作用机制与Cyclin D1及Caspase-3表达有关。
Objective To observe the anti-tumor effect of Qiyi Yigangan decoction on H22 tumor-bearing mice and to explore its mechanism. Methods The right subaxillary subcutaneous tumor caused the solid tumor model. Thirty mice were divided into three groups: control group, TCM group and 5-Fu group. The tumors were weighed and the tumor inhibition rate, thymus index and spleen index were calculated 15 days after treatment. The expressions of CyclinD1 and Caspase-3 were detected by immunohistochemistry. Results Qiye Yigangan decoction inhibited the growth of mouse hepatoma cells by 37.28% and 50.48% in 5-Fu group. The expression of Cyclin D1 and Caspase-3 in all three groups were positive. The statistical results showed that the expression of CyclinD1 and Caspase-3 in traditional Chinese medicine group were significantly decreased compared with the control group. Conclusion Qiyi Yigangan decoction significantly inhibited the proliferation of hepatoma cells, and its mechanism of action was related to the expression of Cyclin D1 and Caspase-3.
