目的研究补阳还五汤(BYHWD)对失神经肌萎缩SD大鼠神经生长因子(NGF)分泌的影响。方法选取雄性SPF级SD大鼠48只,随机分为6组,补阳还五汤高中低剂量组、模型组、假手术组,弥可保组。分别建立腓总神经夹伤模型,手术后每日灌胃,21d后测量足趾宽度,记录展指功能;进行肌电图检测,测量肌电位衰减率;检测胫前肌的湿重比;运用免疫组化法检测大鼠NGF的分泌情况。结果 1展趾功能测定:BYHWD高剂量组、弥可保组与模型组比,显著增高(P<0.01或P<0.05);2电位衰减率:BYHWD高剂量、弥可保组与模型组比较差异显著(P<0.0001或P<0.01)。中药高剂量与中、低剂量组相比较也具备了极显著差异(P<0.0001或P<0.01);3胫前肌湿重比:BYHWD高剂量、弥可保组与模型组相比,均有显著增高(P<0.05);4 NGF蛋白的分泌:假手术组、中药各组和弥可保组可见较多NGF免疫反应阳性颗粒,呈浅棕色。模型组大鼠的NGF阳性颗粒数目减少,着色浅淡。BYHWD高中低剂量组、弥可保组与模型组比差异显著(P<0.01或P<0.05)。结论补阳还五汤能明显降低胫前肌湿重比的退变和电位衰减率,促进展趾功能的恢复,对失神经肌萎缩有很好的治疗效果,其机制可能是通过促进NGF蛋白的分泌实现的。
Objective To investigate the effect of BYHWD on the secretion of nerve growth factor (NGF) in rats with denervated muscular atrophy (SD). Methods Forty-eight male SPF SD rats were randomly divided into 6 groups: Buyang Huanwu Decoction high and low dose group, model group, sham operation group and Mi Kebao group. The common peroneal nerve injury models were established. After operation, the rats were intragastrically administered daily. The toe width was measured after 21 days and the function of the index finger was recorded. The EMG test was performed to measure the myoelectric potential decay rate. The wet weight ratio of the anterior tibialis muscle was measured. Immunohistochemistry was used to detect the secretion of NGF in rats. Results 1 toe function test: BYHWD high-dose group, Mi Kebao group was significantly higher than the model group (P <0.01 or P <0.05); 2 potential decay rate: high BYHWD dose, miconazole group compared with the model group The difference was significant (P <0.0001 or P <0.01). Compared with the medium and low dose groups, the traditional Chinese medicine high dose also had significant difference (P <0.0001 or P <0.01); 3 Compared with the high dosage of BYHWD, (P <0.05); 4 NGF protein secretion: sham operation group, the Chinese medicine group and the Mi Kebao group more NGF immunoreactive granules, light brown. The number of NGF-positive particles in the model group decreased, and the staining was light. BYHWD high school and low dose group, Mi Kebao group and model group than the significant difference (P <0.01 or P <0.05). Conclusion Buyang Huanwu Decoction can significantly reduce the antegrade tibiae wet weight ratio degeneration and potential decay rate and promote the function of the recovery of the toe, have good therapeutic effect on denervated atrophy, its mechanism may be through the promotion of NGF protein The secretion of the realization.
