目的:研究大剂量(2Gy)和低剂量(0.075Gy)X射线诱导pEgr P16重组质粒稳定转染的食管癌细胞株EC9706凋亡的作用。方法:将脂质体包裹的pEgr P16重组质粒,转染食管癌细胞系EC9706中,经G418筛选,获得稳定表达的细胞;采用Westernblot方法检测不同剂量X射线诱导后P16的表达;观察不同剂量X射线照射后,EC9706细胞凋亡率的变化。结果:pEgr P16重组质粒转染EC9706细胞,并获得稳定转染的细胞;2Gy和0.075GyX射线照射均可诱导P16表达增强;稳定转染的细胞经2Gy和0.075GyX射线照射,细胞凋亡率明显高于假照射(0Gy)组(P<0.05~0.01)。结论:0.075GyX射线照射可诱导稳定转染的EC9706细胞中P16表达增强,使诱导细胞凋亡率提高。
AIM: To investigate the effects of high dose (2Gy) and low dose (0.075Gy) X-rays on the apoptosis of EC9706 cells stably transfected with pEgr P16 recombinant plasmid. Methods: The pEgr P16 recombinant plasmid was transfected into esophageal cancer cell line EC9706 and was screened by G418 to obtain stable cells. Western blot was used to detect the expression of P16 after different doses of X-ray irradiation. The effects of different doses of X Changes of apoptosis rate of EC9706 cells after radiation. Results: The recombinant plasmid pEgr P16 was transfected into EC9706 cells and stable transfected cells were obtained. The expression of P16 was induced by 2Gy and 0.075Gy X-ray irradiation. The stably transfected cells were irradiated with 2Gy and 0.075Gy X-ray and the apoptosis rate was significantly Higher than the sham group (0Gy) (P <0.05 ~ 0.01). Conclusion: 0.075Gy X-ray irradiation can induce enhanced expression of P16 in stably transfected EC9706 cells and increase the rate of induced apoptosis.
