目的:观察不同剂量冠心合剂对大鼠急性心肌梗死血流动力学、心梗面积边缘区细胞凋亡及蛋白表达的影响。方法:取雄性SD大鼠80只,随机分为模型组、冠心合剂高剂量组、中剂量组、低剂量组和对照组。手术后4 h测量血流动力学变化后,留取心脏标本,检测梗死面积、细胞凋亡指数以及Bax、Bcl-2、Caspase-3蛋白表达量。结果:与模型组比较,冠心合剂组可明显改善心梗大鼠血流动力学,减少心梗面积及心肌细胞凋亡指数,升高Bcl-2表达,减少Bax和Caspase3蛋白表达。结论:冠心合剂改善心梗大鼠血流动力学,抑制梗死边缘区心肌细胞的凋亡、减少心肌梗死面积,其机制与调节Bcl-2和Bax表达,减少Caspase-3蛋白的表达水平有关。
Objective: To observe the effects of different doses of Guanxin Mixture on hemodynamics, apoptosis and protein expression in the marginal zone of myocardial infarction in rats with acute myocardial infarction. Methods: Eighty male Sprague-Dawley rats were randomly divided into model group, Guanxin Mixture high dose group, middle dose group, low dose group and control group. After 4 hours of operation, the hemodynamic changes were measured and the heart specimens were taken for determination of the infarct size, apoptotic index, and the expressions of Bax, Bcl-2 and Caspase-3 proteins. Results: Compared with the model group, Guanxin Mixture group could obviously improve the hemodynamics, reduce the myocardial infarction area and the myocardial apoptosis index, increase the expression of Bcl-2 and decrease the expression of Bax and Caspase3. Conclusion: Guanxin Mixture can improve the hemodynamics of myocardial infarction rats, inhibit the apoptosis of myocardial cells in infarct border area and reduce the area of myocardial infarction. Its mechanism is related to the regulation of Bcl-2 and Bax expression and the decrease of Caspase-3 protein expression .
