目的探讨辛伐他汀对高脂喂养的 apo E-/-小鼠心肌肥厚及氧化应激分子的影响。方法 5 周龄雄性 apo E-/-小鼠和 C57 小鼠,分为野生型组 (Wild)、apo E-/-小鼠组 (Apo E)及 apo E-/-小鼠+辛伐他汀组 (Apo E-S),予以高脂喂养。8 周龄开始分别给药,干预 3 周后测量舒张期左室前壁厚度 (LVAWd)、舒张期左室后壁厚度 (LVPWd)、心脏体重比 (HW/BW)、左室横截面面积 (LVCSA) 和心肌心房钠尿肽 (ANP) 蛋白表达等心肌肥厚参数及心肌 ROS 水平、p22phox 和 p47phox 蛋白表达等氧化应激指标。结果 Apo E 组 LVAWd 、LVPWd、HW/BW、LVCSA、ANP、ROS、p22phox 及 p47phox 水平均明显高于 Wild 组 (p<0.05),而 Apo E-S 组上述指标均显著低于 Apo E 组 (p<0.05)。结论辛伐他汀可能通过抗氧化应激作用改善高脂饮食条件下 apo E-/-小鼠心肌肥厚。
Objective To investigate the effects of simvastatin on cardiac hypertrophy and oxidative stress in apo E - / - mice fed with high fat diet. Methods Five weeks old male apo E - / - mice and C57 mice were divided into three groups: wild, apo E - / - mice and apo E - / - mice + simvastatin Group (Apo ES), high-fat feeding. The left ventricular anterior wall thickness (LVAWd), diastolic left ventricular posterior wall thickness (LVPWd), cardiac weight ratio (HW / BW), left ventricular cross-sectional area LVCSA) and cardiac atrial natriuretic peptide (ANP) protein expression and myocardial hypertrophy parameters such as myocardial ROS levels, p22phox and p47phox protein expression and other indicators of oxidative stress. Results The levels of LVAWd, LVPWd, HW / BW, LVCSA, ANP, ROS, p22phox and p47phox in Apo E group were significantly higher than those in Wild group (P <0.05), while those in Apo ES group were significantly lower than those in Apo E group 0.05). Conclusion Simvastatin may ameliorate cardiac hypertrophy in apo E - / - mice under high fat diet by antioxidative stress.
