目的观察赤雹根总皂苷(TSTR)对2型糖尿病(T2DM)模型大鼠血糖、血清胰岛素(INS)、胰岛素抵抗指数(IRI)、胰岛素敏感指数(ISI)及糖化血红蛋白水平(GHb)的影响,探讨TSTR对T2DM的治疗作用。方法 Wistar大鼠,采用高糖高脂饲料加链脲佐菌素(STZ)制备T2DM模型,3周血糖>16.7 mmol/L为造模成功,将模型大鼠随机分为模型对照组、TSTR大剂量组(160 mg·kg-1·d-1)、TSTR小剂量组(80 mg·kg-1·d-1)、盐酸二甲双胍片(MHT)100 mg·kg-1·d-1阳性对照组,各给药组分别ig相应药物,模型组ig等体积的蒸馏水。测定首次给药后6 h内血糖及连续给药第3周血糖、INS、及GHb水平并计算IRI和ISI指数。结果首次给药后TSTR大、小剂量组4.6时间点血糖显著低于模型组(P<0.05或P<0.01);连续给药3周,TSTR大、小剂量组血糖、INS、IRI和GHb均显著低于模型对照组(P<0.05或P<0.01);ISI均显著高于模型组(P<0.01)。结论 TSTR对大鼠T2DM有治疗作用,其机制可能与其降低IRI,升高ISI有关。
Objective To observe the effects of TSTR on blood glucose, insulin, insulin resistance index (IRI), insulin sensitivity index (ISI) and glycosylated hemoglobin (GHb) in type 2 diabetes mellitus (T2DM) To investigate the therapeutic effect of TSTR on T2DM. Methods Wistar rats were treated with high-sucrose and high-fat diet supplemented with streptozotocin (STZ) for T2DM. After 3 weeks of blood glucose> 16.7 mmol / L, rats were randomly divided into model group and model group with large TSTR (160 mg · kg -1 · d -1), low dose TSTR group (80 mg · kg -1 · d -1) and metformin hydrochloride tablets (MHT) 100 mg · kg -1 · d -1 Group, the respective drug group ig the corresponding drug, model group ig equal volume of distilled water. The blood glucose levels at 6 h after the first administration and blood glucose, INS, and GHb levels at the third week of continuous administration were measured and IRI and ISI indices were calculated. Results After the first administration of TSTR, the blood glucose of the TSTR large and small dose group was significantly lower than that of the model group at 4.6 (P <0.05 or P <0.01). The blood glucose, INS, IRI and GHb (P <0.05 or P <0.01). ISI was significantly higher than that of the model group (P <0.01). Conclusion TSTR has a therapeutic effect on T2DM in rats, and its mechanism may be related to reducing IRI and increasing ISI.
