目的 :研究化疗药物氟尿嘧啶 (5 - FU)、丝裂霉素 (MMC)、阿霉素 (Dox)、顺铂 (Cis)对人胃癌细胞株 (SGC)的作用特征 ,探讨 SGC细胞株对单一及联合化疗药物的敏感性 ,化疗药物浓度及作用时间对细胞抑制率的影响。方法 :SGC细胞株在含有不同药物的不同浓度的培养基中孵育不同时间 ,用 MTT法测定细胞的抑制率 ,化疗药物浓度参照其在人体的最高峰值浓度 ,选择其峰值浓度的 1/2 5~ 2倍范围 ,作用时间选择 2~ 2 4h。结果 :SGC细胞株对不同化疗药物的敏感性存在差异 ,对Dox最敏感 ,其次为 5 - FU、MMC,对 Cis不敏感。 5 - FU + MMC、5 - FU + Cis及 5 - FU + Dox+ MMC的联合应用表现了较明显的协同作用 ,其中 5 - FU、Dox及 MMC三药联用最显著 ;尽管 Cis单用不敏感 ,在联合应用中也表现了较强的协同作用。相同药物作用时间下 ,随着药物浓度的增加 ,Dox、5 - FU和 MMC对 SGC细胞株的抑制率明显增加 ,Cis的抑制率无明显变化 ;相同浓度下 ,药物作用时间对 SGC细胞株的抑制率也有显著影响 ;化疗药物的联合应用也表现了相似的特征。结论 :SGC细胞株对不同化疗药物的敏感性不同 ,化疗药物的联合应用可明显提高对细胞株的抑制率 ,单用不敏感的药物 ,联合应用中也可能表现较明显的协同作用 ;在影响药效动力学的主要?
