目的观察不同浓度地塞米松对大鼠体外培养脊髓背角星形胶质细胞P2Y1受体表达及TNF-α分泌的影响。方法培养并纯化大鼠脊髓背角星形胶质细胞,地塞米松(终浓度分别为0.1、1、10μmol/L)处理24 h后,采用免疫组织化学染色观察P2Y1受体的表达,双抗夹心间接酶联免疫吸附法测定上清液中TNF-α含量。结果地塞米松(0.1、1、10μmol/L)作用24 h后,与对照组比较,脊髓背角星形胶质细胞P2Y1受体表达下调,并呈剂量依赖性(P<0.01),TNF-α分泌量亦呈剂量依赖性下降(P<0.01)。结论地塞米松能抑制体外培养大鼠脊髓背角星形胶质细胞P2Y1受体表达及TNF-α的分泌。
Objective To observe the effect of dexamethasone on the expression of P2Y1 receptor and the secretion of TNF-α in rat spinal cord astrocytes cultured in vitro. Methods The spinal cord dorsal horn astrocytes and dexamethasone (final concentrations were 0.1, 1, 10 micromol / L) were cultured and purified for 24 h. The expression of P2Y1 receptor was observed by immunohistochemical staining. The contents of TNF-αin supernatant were measured by sandwich indirect enzyme-linked immunosorbent assay. Results After treated with dexamethasone (0.1, 1, 10 μmol / L) for 24 h, the expression of P2Y1 was down-regulated in a dose-dependent manner in spinal dorsal horn astrocytes (P <0.01) α secretion also showed a dose-dependent decline (P <0.01). Conclusion Dexamethasone can inhibit P2Y1 receptor expression and secretion of TNF-α in spinal dorsal horn astrocytes in vitro.
