目的建立测定东方肉穗草中槲皮素含量的方法。方法采用RP-HPLC法,色谱柱为Shimadzu Shimpack VP C18柱(150 mm×46 mm,5μm),流动相为甲醇-0.1%磷酸(45:55),流速为1.0 ml.min-1,检测波长为365 nm,柱温为40℃。结果槲皮素0.025~0.400 mg.ml-1与峰面积呈良好的线性关系,回归方程为:Y=7.74×107X-2.69×105(r=0.9998),平均回收率为99.8%,RSD=0.83%(n=5)。结论所建方法快速、简便,重复性和稳定性均好,能准确地测定东方肉穗草中槲皮素的含量。
Objective To establish a method for the determination of quercetin content in Oriental Safflower. Methods The RP-HPLC method was used. The column was Shimadzu Shimpack VP C18 column (150 mm×46 mm, 5 μm). The mobile phase was methanol-0.1% phosphoric acid (45:55). The flow rate was 1.0 ml.min-1. At 365 nm, the column temperature is 40°C. Results There was a good linear relationship between quercetin 0.025-0.400 mg.ml-1 and the peak area. The regression equation was: Y=7.74×107X-2.69×105(r=0.9998). The average recovery was 99.8%. RSD was 0.83. % (n=5). Conclusion The established method is rapid, simple, reproducible and stable. It can accurately determine the content of quercetin in the oriental taro.
