许多消化和胃肠药都含有蛋白水解酶。通常用酪蛋白-福林法来测定其活性,但该法操作冗繁、费时,不便用作常规质量控制中的测定。本文报道用色素结合型基质——偶氮胶原(Azocoll)来测定胃肠药中蛋白水解酶活性的方法。偶氮胶原为胶原与偶氮色素结合在一起的一种不溶性蛋白质,它与酶反应能生成可溶性有色分解产物,可方便地进行比色测定。该法与传统法相比,大大简化了操作,并有灵敏度高、有色水解产物稳定时间长
Many digestive and gastrointestinal drugs contain proteolytic enzymes. Its activity is usually measured by the casein-Forint method, but this method is cumbersome, time-consuming and inconvenient to use as a measure for routine quality control. Here we report a method for the determination of proteolytic enzyme activity in gastrointestinal drugs by the use of Azocoll, a pigment-binding matrix. Azo-collagen is an insoluble protein that combines collagen and azo pigment. It can react with enzyme to produce soluble colored decomposition products, and can be conveniently used for colorimetric determination. Compared with the traditional method, the method greatly simplifies the operation and has high sensitivity and long stable time of colored hydrolyzate
