The mechanism of immune response to rheumatoid arthritis (RA) remains unclear and may be related to the ongoing development of autologous mixed lymphoid tissue responses to the production of lymphokines in the joints, including the activation of γ-IFN in locally activated macrophages (Mφ). γ-IFN is a potent inducer of surface expression of MHC class II molecules in vitro, and an increase in HLA-DR expression is a characteristic immunohistological feature of rheumatoid arthritis. In order to estimate the inactivation of Mφ by γ-IFN in RA, sensitive natural radioimmunoassay was used to investigate the effects of γ-IFN induced by mitogen and naturally produced by RA lymphocytes.
