目的通过获得的斑蝥抗血清,建立快速、简便、有效的斑蝥素间接竞争ELISA检测方法。方法以合成的斑蝥素完全抗原免疫家兔,获得高效价的抗血清,通过优化条件,建立斑蝥素间接竞争ELISA检测方法的标准曲线,并设定浓度梯度,测定检测灵敏度、检测稳定性和可靠性。结果建立的标准曲线方程为y=-13.421x+102.45,R2=0.988,检测范围为:0.1~24μg/ml。其最低检测下限为0.01μg/ml,批内和批间变异系数分别为4.27%和4.56%。结论本方法具有良好的灵敏性和可重复性,可为进一步开发斑蝥素检测试剂盒提供基础。
OBJECTIVE: To establish a rapid, simple and effective method for the indirect ELISA assay of cantharidin by using the obtained anther serum. Methods Rabbit was immunized with the complete antigen of cantharidin to obtain a high titer of antiserum. By optimizing the conditions, a standard curve of indirect competitive ELISA for cantharidin was established and the concentration gradient was set. The detection sensitivity, stability and reliability were tested Sex. Results The standard curve equation established was y = -13.421x + 102.45, R2 = 0.988, and the detection range was 0.1 ~ 24μg / ml. The lowest limit of detection was 0.01μg / ml. The intra-assay and inter-assay coefficients of variation were 4.27% and 4.56% respectively. Conclusion The method has good sensitivity and repeatability, which can provide a basis for further development of cantharidin detection kit.
