本文报道用低分化胃癌细胞株MKN-46-9直接免疫小鼠制得抗胃癌的单克隆抗体4株即MG5、7、9、11。杂交瘤细胞株已连续培养360天,至今分泌抗体稳定,效价高。抗体能直接用于常规福尔马林固定石蜡包埋的切片染色,不需胰酶消化。经614例ABO免疫组化法染色鉴定,MG对成人正常的胃粘膜、食管粘膜、结肠粘膜以及肺癌、肝癌、胰腺癌、皮肤癌、眼部癌、卵巢癌等癌及其正常组织均呈阴性反应。MG7和MG9两种抗体在胃癌组织中的染色程度和染色阳性细胞数均显著高于结肠癌、食管癌和肺癌(p<0.01)。提示MG7及MG9所识别的抗原主要分布在胃癌组织中。由于这组抗原与CEA等常见肿瘤相关抗原无关,初步认为是一组新的胃癌相关抗原。
In this paper, we report that 4 strains of monoclonal antibodies against gastric cancer, namely MG5, 7, 9, 11 were prepared by immunizing mice with poorly differentiated gastric cancer cell line MKN-46-9. Hybridoma cell lines have been continuously cultured for 360 days, and secreted antibodies have been stable and have high titer so far. The antibodies can be directly used for formalin-fixed paraffin-embedded section staining without trypsin digestion. 614 cases of ABO immunohistochemical staining identified MG negative for normal adult gastric mucosa, esophageal mucosa, colon mucosa, and lung cancer, liver cancer, pancreatic cancer, skin cancer, eye cancer, ovarian cancer and other normal tissues. reaction. The staining of MG7 and MG9 antibodies in gastric cancer tissues and the number of positive cells were significantly higher than those of colon cancer, esophageal cancer, and lung cancer (p<0.01). It is suggested that the antigens recognized by MG7 and MG9 are mainly distributed in gastric cancer tissues. Since this group of antigens is not associated with common tumor-associated antigens such as CEA, it was initially considered to be a new group of gastric cancer-associated antigens.
